Been out of the loop for awhile- a couple questions

For the past several months I've been out of the Roku loop.. Looking to start messing with it again, and have a couple questions:

First off let me say that I really appreciate the hard work that goes into the development here.

What is the established known to work format for DVD rips that will direct play? I have MakeMKV and want to know the ideal settings to use. I see that there are a few different MKV types-  H.264, H.265, H.265ac, etc. Would prefer un-compressed files.

Lastly have there been any developments regarding +/- chapter support without exiting the movie?- like you would on a standard DVD?

Oh, and just to add my Roku3 is ethernet wired.

Does that even matter at this point?  The server will transcode if needed.  You can tune the Roku client bandwidth to the point that something that will play natively will just be streamed.  One of my Roku's is wired and I think it's maxed out on that setting.  But basically, your stuff will just play.  Myself, I tend to use MakeMKV too.  Some things I rip without compression because I want every bit unaltered for my HTPC setup.  Other content may take a trip thru Handbrake if I feel I don't need to store the best quality for a given title.  Those big bluray rips are too much for the Roku, so MB server steps in and transcodes it as needed.  The device just has it's limits but MB makes it work and does a great job at it too.  I can't speak to the chapters thing because I've never been one to use chapters, even on disc.
